Distal-less homeobox 6 (DLX6) is a member of the homeobox transcription factor gene family, highly conserved and similar to the Drosophila distal-less gene. The family comprises at least six genes instrumental in the development of the forebrain, craniofacial structures, limbs, and skeletal formation. In humans, DLX6 regulates enhancer elements responsible for gene expression in neural crest and forebrain derivatives, often working with its paralog DLX5. DLX6 activity is essential for the migration and differentiation of cortical interneurons, craniofacial identity (especially lower jaw patterning), limb development, and osteoblast differentiation for bone formation. Dysregulation, including abnormal antisense RNA (DLX6-AS1), is implicated in a variety of diseases such as neurodevelopmental disorders (often co-acting with DLX5), congenital craniofacial and limb malformations, and cancers. DLX6 is typically not a direct druggable target, but alterations in its gene network may inform disease mechanisms and biomarker development.
