DIX domain-containing protein 1 (DIXDC1) is an intracellular scaffolding protein that contains a DIX (Dishevelled–Axin) domain and serves as a positive regulator of canonical Wnt/β-catenin signaling[1][2][4]. It interacts with partner proteins, including Dishevelled and DISC1, to control cytoskeletal organization, centrosome function, neural progenitor proliferation, neuronal migration, and synaptic structure[1][3][5]. In vasculature, DIXDC1 regulates developmental and pathological angiogenesis by stabilizing VEGFR2 in response to Wnt signaling, facilitating vessel growth and repair under hypoxic conditions[2]. Dysregulation and upregulation of DIXDC1 have been linked to enhanced proliferation, invasion, and metastasis in multiple cancers via PI3K/AKT- and Wnt/β-catenin–dependent pathways[1]. Rare germline variants or loss of DIXDC1 disrupt neurodevelopmental signaling, contributing to increased risk for autism, schizophrenia, and bipolar disorder[3][5]. DIXDC1 is considered an important nodal protein integrating signal transduction pathways relevant to both developmental processes and disease pathogenesis[1][2][3][5].
