DLD-1 is a human colorectal adenocarcinoma cell line established in 1977 from a patient with a Duke's type C colorectal tumor (Dexter et al., 1979; ATCC, CCL-221). It is one of two cell lines, the other being HCT-15, derived from the same tumor specimen, though they exhibit distinct karyotypes and properties. DLD-1 is widely utilized in preclinical oncology research as a model system to study colorectal cancer biology, including cell signaling, metabolism, and the mechanisms of drug resistance. Genetically, the cell line is characterized by several key mutations, including KRAS (G13D), PIK3CA (E545K), and TP53 (C241F), and it displays a microsatellite instability-high (MSI-H) phenotype (Cellosaurus, CVCL_0248). While DLD-1 is frequently used to screen and evaluate the efficacy of chemotherapeutic agents and targeted therapies, it is a biological model rather than a specific molecular target itself. Its use in research helps identify potential therapeutic vulnerabilities and understand the complex interactions within the tumor microenvironment.
