Overview

DLGAP1 antisense RNA 1 is a cytoplasmic long non-coding RNA identified as an oncogenic regulator in multiple human cancers, including gastric cancer, hepatocellular carcinoma, and glioma. Mechanistically, it generally acts as a competing endogenous RNA (ceRNA) or molecular sponge for various tumor-suppressive microRNAs, such as miR-628-5p, miR-515-5p, and miR-26a-5p, lifting repression on oncogenes like AEG-1, DDX59, and ROCK1, and consequently promoting cell proliferation, migration, invasion, and epithelial-mesenchymal transition, while inhibiting apoptosis. Elevated expression of DLGAP1-AS1 is associated with higher tumor grade, advanced stage, metastasis, and poor clinical outcomes across several tumor types. Active research is ongoing to establish DLGAP1-AS1 and its regulatory axis as a potential diagnostic, prognostic, and therapeutic biomarker in oncology.
