Overview

DLX6 antisense RNA 1 (DLX6-AS1), also known as Evf-2 and several other aliases, is a long non-coding RNA (lncRNA) located on human chromosome 7q21.3[5][6]. It is transcribed antisense to the DLX6 gene and has conserved homologs/splicing variants in other mammals[5][1]. DLX6-AS1 is a developmentally regulated lncRNA that plays a critical role in the transcriptional regulation of the DLX5 and DLX6 genes, particularly during brain development and organogenesis[1][5]. It interacts with transcription factors (such as DLX proteins and MECP2), modulating chromatin state, enhancer DNA methylation, and gene expression in neural stem cells and GABAergic interneuron precursors[3][5]. Aberrant upregulation of DLX6-AS1 has been observed in multiple human cancers and is correlated with increased tumor proliferation, invasion, metastasis, and poor prognosis[4][6]. DLX6-AS1 also participates in the regulation of gene expression in non-malignant disorders, such as diabetic nephropathy, where its level reflects disease severity and renal inflammatory status[2]. It is both a potential biomarker and a therapeutic target, although direct drugs targeting this lncRNA are not yet identified, and therapeutic modulation poses substantial developmental and safety challenges due to its essential regulatory roles[4][6].
