Overview

DMP1 and DSPP antisense RNA 1 (DMP1-AS1) is a long non-coding RNA (lncRNA) situated near the DMP1 gene region and known to regulate transcription of the DMP1 gene, which is crucial for dentin and bone mineralization[1][3]. DMP1-AS1 functions at the epigenetic level, modulating H3K27 acetylation in the DMP1 promoter, thereby influencing both DMP1 and the downstream gene DSPP at the mRNA and protein levels. Overexpression of DMP1-AS1 enhances mineralization in osteoblasts, positioning it as an important regulator of skeletal development, though it is not itself a therapeutic target such as a receptor or enzyme[1]. There is evidence that DMP1-AS1 (alias lnc-SPARCL1-1:2) is upregulated in non-alcoholic steatohepatitis (NASH), indicating potential use as a biomarker in metabolic liver disease[2]. There are currently no drugs targeting DMP1-AS1 and its mechanism of action is limited to gene regulation via chromatin modification rather than direct molecular interaction with therapeutic compounds.
