Overview

DNA, proteins, and lipid membranes represent the three fundamental classes of biological macromolecules and structures that constitute living cells. DNA functions as the primary carrier of genetic information, proteins serve as the functional workhorses (enzymes, receptors, transporters), and lipid membranes provide the essential physical barriers and platforms for signaling [PubMed: 27811931]. In pharmacology, these are often targeted collectively by non-specific agents such as alkylating chemotherapeutics, ionizing radiation, or reactive oxygen species (ROS), which induce widespread damage including DNA strand breaks, protein carbonylation, and lipid peroxidation [PubMed: 10693912]. While specific drugs are designed to interact with individual members of these classes, the broad targeting of all three is typically associated with cytotoxic or antiseptic mechanisms. This lack of specificity is a major driver of toxicity in clinical applications, as these agents often cannot distinguish between the molecular components of healthy cells and those of pathogens or malignant cells [PubMed: 33603213]. Consequently, while they are critical therapeutic targets, their collective disruption is a double-edged sword in medicine.
