The major groove is a prominent structural feature of the DNA double helix, specifically referring to the larger of the two grooves that spiral around the molecule. These grooves are created by the geometry and orientation of base pairs within the helical structure, resulting in alternating wide (major) and narrow (minor) grooves along each turn of DNA. The major groove provides greater accessibility for proteins and small molecules compared to the minor groove. This makes it a primary site for sequence-specific binding by transcription factors, regulatory proteins, and enzymes involved in replication or repair. Many drugs and small molecules also target this region via "groove binding," which can modulate gene expression or interfere with essential cellular processes.
