DNA alkylation refers to the covalent addition of alkyl groups to specific atoms within DNA bases or the phosphodiester backbone. This process is typically mediated by electrophilic alkylating agents that react with nucleophilic sites on DNA through SN1 or SN2 mechanisms. Alkylated DNA lesions can distort the double helix structure, block replication and transcription machinery, and induce mutations if not repaired. These effects underlie both the therapeutic action (e.g., in cancer chemotherapy) and carcinogenic potential of many alkylating agents.
