DNA and DNA polymerase complexes are fundamental molecular assemblies responsible for the synthesis of new DNA strands during replication and repair processes. These complexes involve the DNA polymerase enzyme binding to a DNA template and a primer to catalyze the addition of deoxyribonucleotides (NCBI, 2023). In clinical medicine, these complexes are critical targets for treating various conditions, including viral infections and cancer. Antiviral drugs often function as nucleoside analogs that, once phosphorylated, are incorporated into the viral DNA by the polymerase, leading to premature chain termination (StatPearls, 2023). In oncology, inhibitors of these complexes disrupt the rapid DNA replication characteristic of malignant cells, thereby inducing apoptosis (PubMed, 2022). However, the high conservation of polymerase domains across species and cell types necessitates careful drug design to minimize off-target effects on host mitochondrial or nuclear DNA synthesis (Nature Reviews Drug Discovery, 2021).
Inhibition of DNA synthesis through competitive binding with natural dNTPs, incorporation into the DNA strand leading to chain termination, or allosteric inhibition of the polymerase enzyme (StatPearls, 2023; PubMed, 2021).
