Overview

DNA and tubulin are two of the most significant molecular targets in cancer chemotherapy, often addressed simultaneously through dual-targeting agents or combination regimens. DNA (Deoxyribonucleic acid) is the carrier of genetic information, and drugs targeting it typically act by causing structural damage, such as alkylation or intercalation, which prevents replication and transcription (NIH, 2023). Tubulin is a globular protein that polymerizes into microtubules, which are vital for maintaining cell shape, intracellular transport, and forming the mitotic spindle during cell division (StatPearls, 2023). When both are targeted, the cell experiences a dual hit: mitotic arrest due to microtubule disruption and apoptosis triggered by DNA damage. This approach is designed to enhance therapeutic efficacy and circumvent drug resistance mechanisms that often arise when only one pathway is inhibited. While few single-molecule dual inhibitors are currently in widespread clinical use, the concept remains a major focus in medicinal chemistry for developing next-generation antineoplastic agents (Journal of Medicinal Chemistry, 2021).
