DNA-binding protein HU-beta, often referred to as HU-2 or hupB in specific species like Mycobacterium tuberculosis, is a small, basic, dimeric protein that plays a critical role in the structural organization of the bacterial nucleoid. It functions as a histone-like protein, facilitating DNA bending, compaction, and stabilization, which are essential for processes such as replication, recombination, and transcription. By binding non-specifically to double-stranded DNA and with high affinity to distorted DNA structures, HU-beta acts as a global regulator of gene expression, particularly under environmental stress. In pathogenic bacteria, it is often essential for survival and virulence, making it an attractive target for the development of novel antimicrobial agents.
