Overview

DNA-binding protein RFX5 (Regulatory factor X5, RFX5) is a human transcription factor that recognizes the X-box motif in major histocompatibility complex class II (MHC-II) gene promoters, enabling their transcriptional activation. RFX5 is a core component of the RFX complex, essential for the expression of MHC-II genes and adaptive immune function; deficiency or mutation leads to a severe immunodeficiency known as bare lymphocyte syndrome. RFX5 also exhibits cancer relevance, being amplified and overexpressed in hepatocellular carcinoma where it promotes tumor growth (partly via transcriptional activation of YWHAQ/14-3-3 tau) and suppresses apoptosis.
