DNA crosslinking by platinum-based compounds is a critical mechanism underlying the cytotoxic effects of these widely used anticancer drugs. Platinum-DNA adducts form when platinum compounds covalently bind to DNA bases, primarily at the N7 position of guanine and adenine residues. These lesions disrupt essential cellular processes such as replication and transcription, ultimately triggering cell cycle arrest and apoptosis if not repaired.
