A DNA crosslinking agent is a chemically diverse molecule that can covalently bind two or more nucleotides within or between strands of DNA, forming crosslinks. These agents are often bifunctional, possessing two reactive ends that enable them to create covalent bonds either within the same strand (intrastrand) or between opposite strands (interstrand) of double-stranded DNA. The formation of such crosslinks disrupts essential cellular processes like DNA replication and transcription, leading to cytotoxicity and cell death—an effect exploited in cancer chemotherapy.
