Overview

DNA crosslinking agents, specifically those utilizing pyrrolobenzodiazepine (PBD) dimers, are synthetic molecules designed to form covalent bonds between DNA strands. PBD dimers bind to the minor groove of DNA, preferentially at sequences containing guanine bases, and create interstrand crosslinks that disrupt DNA replication and transcription. These agents are primarily used as payloads in antibody-drug conjugates (ADCs) to selectively target and kill cancer cells by inducing cell cycle arrest and apoptosis. Specific examples include SG2057, SG3199 and SG3249.
