Overview

DNA crosslinking at the guanine N7 position refers to the formation of covalent bonds between two nucleotides in DNA, specifically involving the nitrogen atom at position 7 (N7) of guanine bases. This site is highly nucleophilic and exposed in the major groove of double-helical DNA, making it a frequent target for alkylating agents and crosslinking chemicals. Crosslinking at this site can inhibit replication and transcription, leading to cell death, and is exploited in some chemotherapies. While N7-Guanine adducts can serve as sensitive biomarkers for exposure to carcinogens, they are generally considered non-promutagenic due to their instability.
