DNA crosslinking induction refers to the process by which covalent bonds are formed between two nucleotides within DNA, or between DNA and proteins, resulting in crosslinks that can disrupt normal cellular processes. These crosslinks can be classified as intrastrand, interstrand, or DNA-protein crosslinks (DPCs). Induction is driven by endogenous agents (nitrous acid, bifunctional aldehydes, oxidative stress) and exogenous agents (environmental pollutants, tobacco smoke, chemotherapeutic drugs). DNA crosslinking interferes with replication and transcription, leading to genomic instability and potentially cancer or premature aging. Cells employ repair pathways like nucleotide excision repair and specialized proteases to resolve crosslinks.
