Overview

DNA crosslinking via platinum agents refers to the process by which platinum-based chemotherapeutic drugs, such as cisplatin, carboplatin, and oxaliplatin, form covalent bonds between DNA bases. This crosslinking can occur within a single DNA strand (intrastrand) or between two complementary strands (interstrand), and in some cases also involves proteins bound to DNA (DNA-protein crosslinks). These lesions distort the helical structure of DNA, block essential processes such as replication and transcription, and if not repaired efficiently, lead to cell cycle arrest and apoptosis. Tumor cells may develop resistance through enhanced repair capacity or loss/mutation of key repair factors.
