DNA crosslinking via platinum coordination complexes refers to the process by which platinum-based compounds, such as cisplatin and its analogs, form covalent bonds with DNA. These interactions result in various types of crosslinks—primarily intrastrand and interstrand DNA-DNA crosslinks, as well as ternary DNA–protein–platinum complexes. This mechanism is central to the cytotoxic action of many clinically used platinum anticancer drugs. Platinum complexes bind preferentially to the N7 position of guanine bases in DNA. The resulting damage triggers cell cycle arrest and apoptosis.
