Overview

DNA damage repair pathway-deficient cells are cells that have lost or have impaired function in one or more of the cellular mechanisms responsible for detecting and repairing DNA damage. These deficiencies can arise from mutations, deletions, or epigenetic silencing of genes encoding key proteins in the various DNA repair pathways. Such defects compromise genome integrity and are associated with increased cancer risk, progression, and altered responses to therapy. They can be targeted therapeutically through synthetic lethality or by exploiting their increased sensitivity to specific genotoxic agents.
