DNA-directed RNA polymerase II subunit C (POLR2C) is the third-largest subunit of RNA polymerase II, a multi-protein enzyme complex essential for the transcription of protein-coding genes and many noncoding RNAs in eukaryotes. POLR2C forms a heterodimer with POLR2J, establishing a core subassembly crucial for RNA polymerase II stability and assembly. POLR2C contributes to the structure and function of the enzyme, supporting dimerization, promoter clearing, and elongation stages of transcription. Disruption in the RNA polymerase II pathway, including the POLR2C subunit, is implicated in cancer, neurodegeneration, and some genetic diseases, but direct targeting of POLR2C for therapy is not established.
