Overview

DNA-directed RNA polymerase II subunit D (POLR2D), also known as RPB4, is the fourth-largest subunit of the eukaryotic RNA polymerase II complex, responsible for the synthesis of messenger RNA precursors and certain small RNAs from DNA templates. POLR2D forms a heterodimer with RPB7 (POLR2G), which dissociates from the core enzyme under stress or suboptimal conditions and is required for efficient transcript stabilization and proper RNA processing during elongation. Structural studies show POLR2D is vital for RNA recognition, binding nascent RNA near the exit channel, and ensuring transcriptional fidelity. Dysfunction or loss of POLR2D impairs global gene expression and is incompatible with cell viability, with implications for development, disease, and cellular stress response
