DNA-directed RNA polymerase II subunit I (POLR2I) is a protein-coding subunit of RNA polymerase II, located on chromosome 19, and is directly involved in the synthesis of mRNA precursors and various non-coding RNAs in eukaryotic cells[1]. This subunit, which includes conserved zinc finger motifs, forms part of the upper jaw of the polymerase complex, contributing to the DNA binding domain and facilitating the translocation and transcription of DNA templates into RNA[5][1]. As a core part of the 12-subunit RNA polymerase II complex, POLR2I ensures fidelity of transcription and interacts with various transcription regulatory factors, but is not independently a therapeutic target, receptor, or enzyme modulated by drugs[2][3][1]. POLR2I dysfunction or altered expression is implicated in several diseases, mostly due to its fundamental role in gene expression and cell function[1].
