DNA-directed RNA polymerase III subunit C (POLR3C) is a core protein component of the eukaryotic RNA polymerase III complex, which is responsible for the transcription of small, essential RNAs (such as tRNAs and 5S rRNA) vital to cellular protein synthesis and some aspects of innate immunity. POLR3C plays a central structural and functional role in transcription initiation and elongation, forming part of the polymerase core. Mutations or dysregulation of POLR3C and other Pol III subunits are linked to various human diseases, including neurodevelopmental disorders and altered cancer cell phenotype through effects on cellular growth, division, and antiviral response.
