DNA-directed RNA polymerases I, II, and III subunit RPABC1 (POLR2E) encodes the fifth largest subunit shared among the three major eukaryotic DNA-dependent RNA polymerases (I, II, III), all of which are responsible for transcribing cellular DNA into various classes of RNA. This subunit is crucial in the RNA polymerase II complex and forms part of the lower jaw surrounding the central cleft to help guide the incoming DNA template. Its activity includes mRNA synthesis and production of ribosomal and small non-coding RNAs, underlining its fundamental role in gene expression. POLR2E participates in interactions with viral and cellular transactivators, and is present in excess relative to other polymerase subunits. Genetic variation and dysregulation of POLR2E have been associated with disease states such as prostate cancer. No approved drugs act directly and specifically on POLR2E, though some general RNA polymerase inhibitors may act on complexes containing this subunit[1][6][4].
