Overview

DNA fragmentation factor subunit alpha (DFFA) is a regulatory protein encoded by the DFFA gene. It is a key component of the DNA fragmentation factor (DFF) complex, which functions as a heterodimer of a 45-kDa regulatory subunit (DFFA) and a 40-kDa subunit with endonuclease activity (DFFB). DFFA acts as an inhibitor of the caspase-activated DNase (DFFB/CAD) until it is cleaved by caspase-3 during apoptosis. Upon cleavage, DFFA releases DFFB, which then digests chromosomal DNA into nucleosomal fragments, a hallmark of apoptosis. DFFA is therefore central to the execution phase of apoptosis, linking caspase activation to DNA fragmentation and chromatin condensation[1][2][3][8].
