The DNA helicase–primase complex is a multi-protein assembly essential for the replication of double-stranded DNA in various organisms, notably in herpesviruses. This complex coordinates two critical enzymatic activities at the replication fork: unwinding duplex DNA (helicase activity) and synthesizing short RNA primers required for DNA polymerases to initiate synthesis. It is an established antiviral drug target.
