DNA intercalation and free radical generation is a mechanism by which certain molecules interact with DNA, leading to significant biological effects such as cytotoxicity, mutagenesis, or therapeutic outcomes. DNA intercalators insert themselves between adjacent base pairs of the DNA double helix, distorting its structure and inhibiting replication and transcription. Subsequently, these molecules or upon activation by light or cellular enzymes, generate free radicals that cause DNA strand breaks and further cellular damage. This combined action is exploited in many chemotherapeutic agents to selectively damage rapidly proliferating cells' genomes.
