DNA polymerase alpha 2 accessory subunit (POLA2) is a non-catalytic, accessory protein component of the DNA polymerase alpha complex, a key multisubunit enzyme essential for the initiation of eukaryotic DNA replication. POLA2 forms a stable heterodimer with the catalytic subunit (POLA1) and serves as a structural and regulatory scaffold, mediating recruitment of the polymerase–primase complex to replication forks, and facilitating primer synthesis and early DNA strand extension[1][2][4]. POLA2 is crucial for both the initiation of DNA synthesis and for telomere C-strand maintenance. Pathogenic variants in POLA2 can cause developmental disorders with short telomeres, and its malfunction may contribute to broader replication stress-associated diseases[2][4].
