Overview

The POLA1 gene encodes the p180 catalytic subunit of the DNA polymerase alpha (Pol α) complex, essential for initiating eukaryotic DNA replication at origins of replication on both leading and lagging strands. It plays a critical role in synthesizing RNA-DNA primers during Okazaki fragment formation on the lagging strand. The enzyme has limited processivity and lacks 3′ exonuclease activity.
