Overview

DNA polymerase alpha catalytic subunit (POLA1) is the p180 component of the DNA polymerase α-primase complex. It is essential for initiating DNA replication at origins of replication on both leading and lagging strands, and in synthesizing Okazaki fragments on the lagging strand. It works by extending RNA primers with short stretches of DNA before handing off to more processive polymerases. POLA1 interacts with MCM4, GINS1, retinoblastoma protein (RB), PARP1, and RBMS1. Mutations or dysregulation are associated with X-linked reticulate pigmentary disorder and immune dysfunction.
