Overview

DNA polymerase delta (Pol δ) is a multi-subunit enzyme complex found in eukaryotic cells, playing a central role in chromosomal DNA replication, repair, and recombination. It is one of the primary replicative polymerases responsible for synthesizing both the leading and lagging strands during DNA replication. Human Pol δ consists of four subunits: p125 (catalytic; encoded by POLD1), p50 (POLD2), p68 (POLD3), and p12 (POLD4). The enzyme’s processivity is greatly enhanced by its interaction with proliferating cell nuclear antigen (PCNA). Beyond replication, Pol δ participates in various repair pathways. Improper regulation or mutation within any component/subunit can disrupt normal cell cycle progression or damage response pathways, and defects have been linked with tumorigenesis.
