DNA polymerase delta subunit 2 (POLD2) is the 50-kDa accessory subunit of the DNA polymerase delta complex, which is essential for high-fidelity DNA replication and repair. As a non-catalytic component, POLD2 stabilizes the holoenzyme, promotes processivity through interaction with the proliferating cell nuclear antigen (PCNA), and is involved in both lagging and leading strand synthesis. It is also a component of the DNA polymerase zeta complex, supporting translesion synthesis and various DNA repair pathways, including nucleotide excision repair. Mutations or altered expression of POLD2 are linked with increased genomic instability and cancers.
