DNA polymerase III PolC is a replicative DNA polymerase found in many Gram-positive bacteria. It is essential for genome duplication and is the primary enzyme responsible for high-fidelity, processive DNA synthesis during bacterial cell division. The enzyme also exhibits 3' to 5' exonuclease activity, which provides proofreading capability to ensure accurate DNA replication. Its unique structural features make it an attractive target for antibacterial drug development because inhibiting PolC can halt bacterial proliferation without affecting eukaryotic host cells.
