DNA polymerase theta is an error-prone eukaryotic enzyme encoded by the *POLQ* gene that plays an essential role in repairing double-strand breaks through the mutagenic TMEJ/MMEJ pathway. Its unique structure combines a helicase-like domain with an A-family polymerase lacking proofreading ability. While crucial for genome stability under stress conditions, its overexpression supports tumor cell survival when canonical DSB pathways fail, making it both a biomarker of poor prognosis and an emerging therapeutic target—especially relevant where homologous recombination defects exist.
