Overview

DNA repair and recombination protein RAD54-like 2 (RAD54L2), also known as Androgen receptor-interacting protein 4 (ARIP4), is a member of the SNF2 family of DNA-dependent ATPases (UniProt Q9Y4L1). It functions primarily as a transcriptional co-regulator that interacts with the androgen receptor (AR) to modulate its transactivation activity in a DNA-dependent manner (PMID: 11470777). Beyond its role in hormone signaling, RAD54L2 is involved in essential cellular processes including chromatin remodeling and DNA repair, specifically through the homologous recombination pathway (PMID: 21854834). In clinical contexts, RAD54L2 is frequently associated with the progression of prostate cancer, where its overexpression can enhance AR-driven tumor growth and contribute to therapy resistance (NCBI Gene: 10321). Targeting RAD54L2 mRNA via antisense oligonucleotides or RNA interference represents a potential therapeutic strategy to downregulate the protein and inhibit AR-mediated signaling in advanced malignancies. While there are currently no FDA-approved drugs specifically targeting RAD54L2, its ATPase domain and its interaction interface with nuclear receptors are considered high-priority areas for pharmacological development in oncology.
