Overview

DNA replication and sister chromatid cohesion 1 (DSCC1) is a protein that is a critical component of the alternative replication factor C (RFC) complex known as the CTF18-RFC complex, which includes CTF18, DSCC1, and CTF8. This complex is responsible for loading the proliferating cell nuclear antigen (PCNA) onto DNA during the S phase of the cell cycle. DSCC1's main functions are to support proper DNA replication, regulate replication fork progression, and ensure the establishment of sister chromatid cohesion necessary for accurate chromosome segregation. DSCC1 is overexpressed in several cancer types, where it promotes cell proliferation, tumor growth, and resistance to apoptosis, making it a potential prognostic biomarker and therapeutic target. Gene amplification and high expression levels of DSCC1 correlate with adverse prognosis in liver, breast, and colorectal cancers. No approved drugs are known to specifically target DSCC1 at this time.
