DNA synthesis (chain termination) is a process utilized to halt DNA strand elongation during replication or sequencing. It relies on the use of dideoxynucleotides (ddNTPs), which lack a 3'-hydroxyl group, causing termination of the DNA chain upon incorporation by DNA polymerase.
