The DNA synthesis enzyme machinery, or replisome, is a complex of proteins and enzymes responsible for accurately replicating DNA during cell division. Key components include DNA polymerases (Pol α, Pol δ, Pol ε), primase, helicase (CMG complex), single-stranded binding proteins (SSB), clamp loader complex (RFC), sliding clamp (PCNA), topoisomerase, and DNA ligase. The replisome ensures faithful duplication of genetic material, and its activity is tightly regulated during the cell cycle. It is a therapeutic target in cancer and viral infections.
