DNA Synthesis Inhibition via DNA Crosslinking (N/A)

Target
N/A
Molecular classification
DNA, Genomic DNA
01

Overview

Platinum-based chemotherapeutic agents inhibit DNA synthesis by forming DNA crosslinks. These crosslinks disrupt DNA replication and transcription, leading to cell cycle arrest and apoptosis. The effectiveness of platinum agents depends on their ability to induce persistent blocks in replication and transcription through stable DNA adduct formation. Resistance mechanisms involving DNA repair pathways are a significant clinical challenge.
