DNA synthesis inhibition via nucleoside analog incorporation is a mechanism by which certain drugs disrupt the replication of DNA in cells. Nucleoside analogs are structurally similar to natural nucleosides but with critical modifications that interfere with normal DNA chain elongation. These analogs are incorporated into newly synthesized DNA strands, causing chain termination and ultimately leading to cell cycle arrest and apoptosis. This mechanism is exploited in antiviral and anticancer therapies.
