DNA topoisomerase 2-binding protein 1 (TOPBP1) is a multi-domain scaffold protein essential for coordinating protein complexes involved in DNA replication, DNA repair, and cell cycle checkpoint activation[2][3][1]. It is characterized by the presence of nine BRCT domains, which facilitate phosphorylation-dependent interactions with numerous proteins, including DNA topoisomerase II, BRCA1, RAD9, MDC1, and ATR[1][2]. TOPBP1 is crucial for activation of the ATR kinase in response to DNA damage, bridging accessory factors and promoting recruitment of repair proteins to damage sites[3][2]. Through its regulatory and structural roles, TOPBP1 is central to genome integrity. Abnormal expression or mutations in TOPBP1 are linked to cancer and other disease states[2].
