Overview

DNA topoisomerase I pseudogene 1 (TOP1P1) is a processed pseudogene derived from the active DNA topoisomerase I (TOP1) gene and localized to chromosome 1[3]. Unlike the functional TOP1 protein that regulates DNA topology during transcription, replication, and repair[1][7], TOP1P1 does not encode an active enzyme. It contains regions homologous to the C-terminal coding sequence of TOP1 and may be transcribed into antisense RNA, potentially influencing regulation of the parental gene, but its biological function remains uncertain[2][3]. There are no known direct links to disease roles, therapeutic drugs, or mechanisms of action for this pseudogene. Pseudogenes can show disease- and tissue-specific expression[5], and some pseudogene RNAs have been implicated in regulatory functions or as biomarkers in cancer, but such roles for DNA topoisomerase I pseudogene 1 have not yet been defined. The designation of TOP1P1 as a pseudogene means it is not considered a therapeutic target[2][3].
