DNA Topoisomerase II Beta (TOP2B) is a type II topoisomerase enzyme essential for managing DNA topology during transcription, replication, chromosome condensation, and chromatid separation. It introduces transient double-stranded breaks in DNA to relieve torsional stress and resolve catenanes. TOP2B is a target for anticancer agents, but mutations can lead to drug resistance and other disease states.
Drugs targeting TOP2B, such as mitoxantrone, induce double-strand breaks in DNA by interfering with the enzyme's ability to religate the DNA after strand passage.
