Overview

DNAJA4 is a member of the Hsp40/DNAJ family of molecular chaperones, specifically classified as DnaJ heat shock protein family (Hsp40) member A4 (gene symbol DNAJA4)[3][7]. It is primarily involved in the regulation of protein folding, refolding, and cellular stress responses, operating in association with the Hsp70 system[5][7]. DNAJA4 is present in the cytosol and associated with cell membranes, and is thought to contribute to protein homeostasis, influence viral replication, and participate in various cellular pathways, including cholesterol biosynthesis and regulation of cell motility[1][7]. High expression of DNAJA4 has been linked to poor survival in breast cancer, while in other cancers (such as nasopharyngeal carcinoma and melanoma), it may act as a metastasis suppressor[2][4]. DNAJA4 is not currently considered a direct drug target but is of interest as a prognostic or predictive biomarker in select cancers due to its differential expression and regulatory roles.
