Overview

DnaJ heat shock protein family member C18 (DNAJC18) is part of the Hsp40/DnaJ family, specifically the type III subfamily, distinguished by a J domain but lacking the typical G/F-rich or cysteine-rich regions found in other subfamilies[2]. It encodes a cytosolic protein, functioning primarily as a chaperone or co-chaperone involved in cellular response to misfolded proteins and maintenance of proteostasis[1][4][7]. It interacts with Hsp70 proteins to regulate their ATPase activity and is suggested to participate in processes such as cellular stress response, protein folding, and possibly in germ cell development (spermatogenesis), particularly in the testis[2]. DNAJC18 is located in the endoplasmic reticulum membrane and cytoplasm, and may play a role in viral infection pathways, notably aiding the penetration of certain viruses, such as polyomavirus, across the ER membrane[1][2][4].
