DnaJ heat shock protein family member C25 (DNAJC25) is a member of the HSP40 (DnaJ) co-chaperone family, primarily functioning in the maintenance of protein homeostasis by partnering with Hsp70 for proper protein folding, especially under cellular stress conditions[2][5]. It is predicted to be membrane-bound and active at the endoplasmic reticulum membrane[2][7]. DNAJC25 is highly expressed in the liver and has been shown to be significantly downregulated in hepatocellular carcinoma, where its overexpression promotes apoptosis and suppresses cell proliferation, supporting a role as a candidate tumor suppressor in liver cancer[4]. The broader HSP40 family, which includes DNAJC25, is implicated in regulating protein quality control, cancer biology, and adaptation to environmental (thermal) stress. There are currently no known drug interactions or specific mechanisms of pharmacological modulation described for DNAJC25[2][4][5].
