Overview

DNAX accessory molecule-1 ligands (DNAM-1 ligands), primarily poliovirus receptor (PVR; CD155) and Nectin-2 (CD112), are cell surface glycoproteins belonging to the nectin and nectin-like families. They are expressed on endothelial cells at intercellular junctions, tumor cells, infected cells, and various normal cells including neuronal, epithelial, and fibroblastic cells. These ligands engage the activating receptor DNAM-1 (CD226) on NK cells, T cells, and monocytes to promote cytotoxicity, cytokine secretion, immune synapse formation, cell adhesion, and monocyte transendothelial migration during diapedesis. PVR (CD155) is the major ligand on endothelial cells and critical for NK-mediated tumor killing, while both ligands contribute to immune recognition of transformed or stressed cells. Expression is upregulated in tumorigenesis, viral infections, and inflammation.[1][2][3]
