Docking protein 5 (DOK5) is a member of the DOK family of membrane-associated adapter proteins, which function in signal transduction pathways by providing a molecular scaffold for assembling multi-protein signaling complexes. DOK5 interacts with phosphorylated receptor tyrosine kinases and mediates neurite outgrowth as well as activation of the MAP kinase pathway, playing a key role in nervous system development and cell signaling. It is notable for not interacting with RASGAP, distinguishing it from other DOK family members. DOK5 is up-regulated in systemic sclerosis and is associated with insulin-like growth factor binding protein 5-induced fibrosis. It is also involved in the regulation of osteoblast proliferation and differentiation via the canonical Wnt/beta-catenin signaling pathway[1][2][3][6]. The gene is located on human chromosome 20. There are no direct drugs known to target DOK5 as a primary therapeutic target, nor are there established mechanisms of action or safety concerns associated with drug targeting of DOK5 at this time[2][3].
